We are seeking a motivated Postdoctoral Researcher to join our team developing an advanced technology platform for cell engineering and cell therapies within the P4Health project. The successful candidate will contribute to the development and implementation of methods for the modification, characterization, and functional assessment of cells for research and translational applications, with a particular focus on developing novel approaches in cell therapy and immunotherapy.
The main responsibilities of the Postdoctoral Researcher will include:
- contributing to the establishment and development of a technology platform for cell engineering and cell therapies;
- developing, optimizing, and validating methods for the genetic modification of cells, including CRISPR/Cas-based approaches and cell transfection/transduction;
- maintaining cell cultures and developing methods for cell expansion and modification;
- developing methods for the functional characterization of cells, including proliferation, viability, cytotoxicity, and other functional assays;
- designing and performing analyses using flow cytometry and multiparametric cell phenotyping;
- applying molecular biology methods to characterize modified cells, including PCR, RT-qPCR and WB for DNA/RNA and protein analyses;
- contributing to the development of imaging methods and quantitative cell analysis;
- designing experiments, analyzing and interpreting results, and preparing experimental documentation;
- developing and standardizing laboratory procedures and protocols (**SOPs**);
- contributing to the development of quality control systems and criteria for the assessment of cellular materials;
- collaborating with teams working in immunology, cancer biology, biobanking, genomics, and advanced therapy development;
- participating in research and development projects conducted in collaboration with academic, clinical, and industry partners;
- preparing scientific publications and reports and presenting research findings at scientific conferences;
- contributing to the development of new research projects and grant applications.

- PhD in Neuroscience, Neurobiology, Physiology, Biomedical Engineering or a related discipline.

You will join the international consortium “DECIPHER: Deciphering the role of CD90+ stromal cells in neuropathic pain”, funded under the ERA-NET programme. The consortium aims to uncover the cellular and molecular mechanisms underlying chronic neuropathic pain and to establish the foundations for novel targeted analgesic therapies. The project is based on the hypothesis that CD90+ stromal cells play a central role in driving and maintaining peripheral sensory neuron hyperexcitability, thereby contributing to persistent neuropathic pain independently of classical inflammation. Your work will focus on the preclinical validation of newly identified therapeutic targets using complementary mouse models of neuropathic pain, integrating behavioural neuroscience with advanced in vivo neurophysiology (pharmacological interventions and functional calcium imaging and multi-electrode arrays, MEA).
You will specialise in advanced in vivo neurophysiology, combining behavioural neuroscience with functional optical imaging of sensory neurons. Using viral delivery of genetically encoded calcium indicators (AAV9-GCaMP6s), you will perform large-scale in vivo calcium imaging of dorsal root ganglion (L3/L4 DRG) neurons together with quantitative behavioural assessment of neuropathic pain. The project combines state-of-the-art optical imaging, pharmacological intervention, computational data analysis and translational neuroscience in close collaboration with leading European laboratories.
The successful candidate will be employed full-time for 36 months within an internationally funded collaborative project and will have the opportunity to work with leading European groups in translational pain neuroscience while developing expertise in advanced systems neuroscience and preclinical therapeutic validation.
You will be responsible for the experimental implementation of the preclinical work package (WP3). Your responsibilities will include:
- performing regulated animal procedures and inducing mouse models of neuropathic pain,
- carrying out behavioural phenotyping (mechanical and thermal hypersensitivity assays together with automated home-ge monitoring),
- performing terminal in vivocalcium imaging experiments from dorsal root ganglion neurons,
- administering and evaluating novel therapeutic compounds, including monoclonal antibodies and receptor ligands,
- analysing large-scale neuronal population imaging datasets using computational pipelines,
- maintaining high-quality experimental documentation and metadata,
- presenting results during consortium meetings and international conferences,
- contributing to manuscript preparation and dissemination of project results.

- Holds or be near completion of PhD in Neuroscience or related discipline (i.e. Neurobiology, Biophysics, etc.),
- Advanced spoken and written English skills,
- Proven research track record,
- Ability to travel and present data internationally,
- Ability to plan your own work and/or the work of the person you line manage (i.e. PhD students),
- Documented work experience with mouse models >1 year (i.e. behavioural tests on mice),
- Good understanding of molecular genetics and viral vectors,
- Good analytical skills.
- Experience with pain models,
- Experience in animal surgery (stereotaxic injections),
- Experience in mouse behavioural assays (affective, locomotor and nociceptive thresholds assessment, preferably with computer vision tools),
- Experience in neurophysiological techniques, especially in vivo functional optical imaging (1-/2- photon)
- Coding skills, or ability to implement written scripts in R/Python/Matlab for data analysis.
The role:
You will work on a generously funded project that delves into presynaptic control of primary sensory neurons by top-down brainstem noradrenergic neurons. You will use novel sensory neuron type specific alpha-2c-adrenoceptor knock-out mice in conjunction with selective optogenetic modulation of the top-down noradrenergic circuits to probe the noradrenergic mechanisms controlling nociception.
You will be specialising in in-depth histological and functional analysis of the knock-out mouse model. An ideal candidate will have a background in transgenic animals work and cutting-edge histological approaches (incl. RNAscope, tissue clearing and expansion microscopy apart from classic immunohistochemistry) with contemporary computational approaches in image analysis. A strong drive to learn and perform in vivo neurophysiological techniques (such as one and 2-photon in vivo calcium imaging) as well as rodent nociceptive behavioural testing will be of advantage

Your main duties:
- you will be to perform in-depth tissue analysis post-mortem.
- You will supervise correct transgenic animal lines breeding (in continuous cooperation with the lab technician) and take responsibility for regulated animal procedures.
- You will ensure that data (and associated metadata) are of the highest quality and in a form that allows for further processing.
You will be asked to:
- independently undertake the research project, and execute experiments in consultation with the group leader,
- participate in lab activities, including group meetings, journal clubs, seminars and conferences,
- write up findings for publications and completing revisions, iv) collaborate with other lab members on daily lab work.
